Cloning and screening bioconversion activities

The biocatalysis activities are mainly based on the construction of collections of enzymes that are representative of the genetic diversity of the families of interest. Selection using bioinformatics tools enables identification of representatives of the diversity, which are cloned in an expression vector. A cloning and high-throughput screening platform exploits a collection of over 1000 prokaryotes. The enzyme collections are then tested using high-throughput screening with natural or non-natural molecules. The idea is to exploit the promiscuity of the enzymes and identify those able to catalyze, even in a marginal manner, an unusual conversion to produce molecules of interest. Currently, our collections contain 5000 enzymes covering the major enzyme families:  hydrolases (lipases, nitrilases), lyases (decarboxylases), oxidases (Baeyer-Villigerases, dehydrogenases, dioxygenases), ligases (aldolases, transketolases), transferases (transaminases, kinases). The platform also contributes to projects related to the discovery of gene functions in their metabolic context in cooperation with the LGBM.​
